In a landmark achievement that oncologists are calling a turning point in cancer treatment, Moderna and Merck announced on August 19, 2026, that their combined mRNA-based cancer vaccine significantly reduced melanoma recurrence and spread in a late-stage clinical trial.
The Phase 3 INTerpath-001 trial enrolled 1,137 patients with high-risk melanoma (Stage IIB through IV) whose tumors had been surgically removed. Participants received either Merck’s Keytruda (pembrolizumab) plus intismeran, Moderna’s personalized mRNA therapeutic, or Keytruda alone. An interim analysis showed the combination met both its primary goal of improving recurrence-free survival and a key secondary goal of preventing distant metastasis.
“This is the first time that we’ve seen really clinically significant, statistically significant improvements over checkpoints like Keytruda in this population, and it is the first time we’ve done that with an individualized treatment,” said Moderna President Stephen Hoge in an interview.
Earlier Phase 2b results, presented at the 2026 ASCO Annual Meeting, showed the combination reduced the risk of recurrence or death by 49% after five years. The Phase 3 trial also demonstrated a 59% reduction in distant metastasis or death risk. No new safety signals emerged, with side effects consistent with prior studies.
Melanoma, the deadliest form of skin cancer, kills approximately 8,500 Americans each year. About 112,000 new cases are expected in the U.S. in 2026 alone. The personalized vaccine works by analyzing mutations unique to a patient’s tumor and creating an mRNA sequence that trains the immune system to recognize and attack cancer cells.
The success has sent shockwaves through the biotech industry. Moderna shares surged as much as 160% on August 19, while Merck stock rose 12%. Analysts project the therapy could generate billions in revenue by 2035. The companies plan to present full results at an upcoming medical meeting and submit for FDA approval within months.
“Today’s results represent a landmark moment for adjuvant melanoma treatment. This is the first Phase 3 study to show that intismeran, a treatment designed based on the unique mutational fingerprint of a patient’s own tumor, given in combination with pembrolizumab can reduce the risk of recurrence or death,” said Professor Georgina Long, the study’s principal investigator at Melanoma Institute Australia.
The breakthrough also marks a remarkable reversal for Moderna, which faced setbacks when the Trump administration canceled contracts for its mRNA programs earlier this year. Investors had grown skeptical about the company’s ability to develop products beyond its COVID-19 vaccine. The cancer vaccine results may accelerate mRNA research across other cancer types now in early testing.
